N2 - The steroid hormone metabolites in urine from adrenal- and gonadectomized female rats have been studied by gas chromatography-mass spectrometry. The C19 (3α, 7α-dihydroxy-5α-androstan-17-one, 3α, 19-dihydroxy-5α-androstan-17-one, 5α-androstane-3α, 7α, 17β-triol, and 3α, 11β-dihydroxy-5α-androstan-17-one), C21O2 (5α-pregnane-3α, 20β-diol) and C21O3 (3α, 19-dihydroxy-5α, 17α-pregnan-20-one, 3α, 17α-dihydroxy-5α, 17β-pregnan-20-one, 3α, 15α-dihydroxy-5α, 17β-pregnan-20-one, and 3α,16α-dihydroxy-5α, 17β-pregnan-20-one) steroid monosulphates in urine from female rats were of ovarian origin, since their excretion was not affected by adrenalectomy, whereas ovariectomy resulted in their disappearance from urine. On the other hand treatment of ovariectomized female rats with the 3β-hydroxy-Δ5-steroid oxidoreductase inhibitor 2α-cyano-4,4,17α-trimethyl-5-androsten-17β-ol-3-one resulted in the excretion of monosulphurylated 3β-hydroxy-5-androsten-17-one, 3β-hydroxy-5-pregnen-20-one, 3α, 16α-dihydroxy-5α, 17β-pregnan-20-one and 3β, 16α-dihydroxy-5-pregnen-20-one in urine. This shows that under abnormal conditions the adrenal can secrete C19 and 21-deoxy-C21 steroids, the metabolites of which are excreted in the urine.
